2. Summary

Dengue viruses, transmitted by Aedes aegypti and Aedes albopictus mosquitoes, are widely distributed throughout the tropical and subtropical areas of the world. There are four known distinct serotypes (dengue 1, 2, 3 and 4). In children, infection is often subclinical or causes a self-limited febrile disease. However, if the patient is infected subsequent times with a different serotype, a more severe disease, dengue hemorrhagic fever or dengue shock syndrome, is more likely to occur. Dengue is considered to be the most important arthropod-borne viral disease due to the  human morbidity and mortality it causes. NS1 is a highly-conserved glycoprotein that is present at high concentrations in the sera of dengue-infected patients during the early clinical phase of the disease.

Dengue NS1 antigen is found from the first day and up to 9 days after onset of fever in sample of primary or secondary dengue infected patients. Immune response includes IgM antibodies produced by the 3rd-5th day of symptoms and persist for 30-60 days. IgG antibodies appear near the 14th day and persist for life. Secondary infections often result in high fever and in many cases with hemorrhagic events and circulatory failure. Secondary infections show that IgG levels rise within 1-2 days after the onset of symptoms and induce an IgM response after 20 days of infection.

4. Assay Procedure

Allow the test device, specimen, buffer, and/or controls to reach room temperature (15-30°C) prior to testing.

(1) Bring the pouch to room temperature before opening. Remove the test device from the sealed pouch and use it as soon as possible.

(2)  Place the test device on a clean and level surface.

(3) Dengue IgM/IgG Rapid Test

For Serum or Plasma Specimens:

Hold the dropper vertically, draw the specimen up to the Fill Line (approximately 5 μL), and transfer the specimen to the specimen well (S) of the test device, then add 2 drops of buffer and start the timer. See illustration below. Avoid trapping air bubbles in the specimen well (S).

For Whole Blood Specimens:

To use a dropper: Hold the dropper vertically, draw the specimen 0.5-1 cm above the Fill Line, and

transfer 1 drop of whole blood to the specimen well (S) of the test device, then add 2 drops of buffer

and start the timer. See illustration below.

(4) Dengue NS1 Antigen Rapid Test

For Serum or Plasma Specimens: 

Hold the dropper vertically  and transfer 3 drops of serum or plasma to the specimen well (S) of the test device, then add 1 drop of buffer and start the timer. See illustration below. 

For Whole Blood Specimens: 

Hold the dropper vertically and transfer 3 drops  of whole blood to the specimen well (S) of the test device, then add 1 drops of buffer and start the timer. See illustration below.

(5) Wait for the colored line(s) to appear. Read results at 15 minutes. Do not interpret the result after 20 minutes.
